I'm using the nptl flag.
But I can't compare as I never used gentoo + audio-application without
this flag :)
So wait for other answers

Hi,
I've used nptlonly for at least 18 months. I've never had a problem
with it as far as I know.
I've never heard of LD_ASSUME_KERNEL so either it's buried in there
somewhere I don't know about or it's not necessary.
